ExperimentCoulterCount

ExperimentCoulterCount[Samples]Protocol

generates a Protocol object for counting and sizing particles (typically cells) of difference sizes in the provided Samples by suspending them in a conductive electrolyte solution, pumping them through an aperture, and measuring the corresponding electrical resistance change caused by particles in place of the ions passing through the aperture. The electrical resistance change is measured by a voltage pulse recorded by the electronics such that the particle count is derived from the number of voltage pulses and the particle size is derived from the pulse shape and peak intensities.

    
ExperimentCoulterCount uses the Coulter principle to count cell numbers and measure cell sizes which is important for monitoring cell health and proliferation rate, seeding cells for subsequent experiments, transfection or infection, and preparing for cell-based assays. The sample with particles (typically cells) is first suspended into an electrolyte solution. A specialized tube, that has a small aperture with electrodes located both inside and outside the tube to form a closed circuit, is then submerged into the electrolyte solution. The integrated pump pumps the cells through the aperture. As each cell passes through the aperture, replacing the ion flow of electrolyte, the effective electrical resistance between two electrodes changes, which is measured by a voltage pulse recorded by the electronics. The number of voltage pulses is interpreted as the particle counts, while the particle size is derived from the pulse shape and peak intensities. For instance, when diagnosing a variety of diseases, red blood cell counts (RBCs) and white blood cell counts (WBCs) can be quickly measured using a 70 µm aperture tube.
    

Experimental Principles

Step 1: Aperture tube (a specialized tube that has a small aperture at the bottom, determined by the "ApertureTube" option) is installed and system is flushed with the electrolyte solution. Step 2 (optional): Sizing accuracy is checked with size standard samples (determined by the options in "System Suitability Check" category). If the system fails the first suitability check, recalibration of the ApertureTube will be triggered. Afterwards, the system suitability check will automatically rerun. If the system fails again, SystemSuitabilityError will be set to True in the protocol object. Step 5: System is flushed with clean ElectrolyteSolution from the instrument reservoir before measurement. Step 6-8: A constant current is applied between inside and outside of the ApertureTube. Integrated pump pumps the SamplesIn-ElectrolyteSolution mixture into the ApertureTube. Each particle passage event is recorded by the voltage changes. Step 9: System is flushed with clean ElectrolyteSolution from instrument reservoir after measurement. Step 10: The voltage data is processed to give information about the size distribution and particle count of the SamplesIn.

Experiment Options

    General

    Instrument

    The instrument that is used to count and size particles in a sample by suspending them in a conductive electrolyte solution, pumping them through an aperture, and measuring the corresponding electrical resistance change caused by particles in place of the ions passing through the aperture. The electrical resistance change is measured by a voltage pulse recorded by the electronics such that the particle count is derived from the number of voltage pulses and the particle size is derived from the pulse shape and peak intensities (see Figure 1.3). Note that the resistivity of the particles can be either larger or less than that of the electrolyte solution.

    ApertureTube

    A glass tube with a small aperture near the bottom through which particles are pumped to perturb the electrical resistance within the aperture for particle sizing and counting. The diameter of the aperture used for the electrical resistance measurement dictates the accessible window for particle size measurement, which is generally 2-80% of the ApertureDiameter.

    ApertureDiameter

    The desired diameter of the aperture used for the electrical resistance measurement, which dictates the accessible window for particle size measurement, which is generally 2-80% of the ApertureDiameter.

    Default Calculation: Automatically set based on the choice of ApertureTube and type of particles in the SamplesIn. Specifically, set to the ApertureDiameter value of the ApertureTube if specified, or set to a value in CoulterCounterApertureDiameterP that is greater than the largest diameters of all compositions in the SamplesIn. If none of the composition has a defined Diameter, set to 100. micrometers to avoid blockage.

    ElectrolyteSolution

    The conductive solution used to suspend the particles to be counted and sized. The electrolyte solution generally contains an aqueous or organic solvent and an electrolyte chemical to make the solution conductive. Please choose a sample solution with a solvent that does not dissolve or damage the target analyte particles to be counted and sized and an electrolyte chemical that maximizes the conductivity of the overall solution. For ApertureDiameter less than or equal to 30 Micrometer, please choose an electrolyte stock solution in the catalog that is filtered with FilterPoreSize less than or equal to 2% of the ApertureDiameter within two days.
    Default Value: Automatic
    Default Calculation: Automatically set to Model[Sample,"Isoton II"] if ApertureDiameter is greater than 30 Micrometer. Otherwise set to Model[Sample,StockSolution,"Beckman Coulter ISOTON II Electrolyte Diluent, 500 mL, filtered"].

    Flushing

    ElectrolyteSolutionVolume

    The amount of the electrolyte solution to be added into the ElectrolyteSolutionContainer of the coulter counter instrument. The amount added here supplies a reservoir of clean electrolyte solution that is pumped to flush the aperture tube before and after sample runs to remove particles that may remain trapped in the bottom of the aperture tube. The minimum amount needed to avoid drawing air into the system is 500 Milliliter.
    Default Value: Automatic
    Default Calculation: Automatically set to the calculated volume to fulfill all the flushes before and after sample runs based on number of SamplesIn plus a 1-Liter buffering volume for ElectrolyteContainer.
    Pattern Description: Greater than or equal to 0 milliliters and less than or equal to 4 liters.

    FlushFlowRate

    The target volume of the electrolyte solution pumped through the aperture of the ApertureTube per unit time when flushing the aperture tube (see Figure 2.1.1).
    Default Value: Automatic
    Default Calculation: Automatically set based on ApertureDiameter and internal PressureToFlowRateStandardCurve standard curve stored in ApertureTube object. If ApertureDiameter is greater than 560 Micrometer, set to a flow rate that maintains the internal pressure during pumping to be 3 Torr. Otherwise, set to a value that maintains the internal pressure to be 6 Torr during pumping.
    Pattern Description: Greater than or equal to 0 microliters per second and less than or equal to 10 milliliters per second.

    FlushTime

    The duration that electrolyte solution flow from ElectrolyteSolutionContainer, to ApertureTube, to ParticleTrapContainer, and to InternalWasteContainer is maintained, in an attempt to remove the particles inside the ApertureTube after connecting the ApertureTube to instrument for the first time, before and after sample runs, or before changing the sample in the MeasurementContainer (see Figure 2.1.1).
    Default Value: 12 seconds
    Pattern Description: Greater than or equal to 0 seconds and less than or equal to 5 minutes.

    System Suitability Check

    SystemSuitabilityCheck

    Indicates if a system suitability check with a particle size standard sample is run prior to the actual sample runs. During system suitability check, one or more size standard samples with known mean diameters or volumes are loaded into the instrument and sized via electrical resistance measurement individually. The system is considered suitable when the measured mean diameter matches the manufacture-labelled mean diameter or the measured mean volume matches the manufacture-labelled mean volume within SystemSuitabilityTolerance. A discrepancy above SystemSuitabilityTolerance triggers the size-calibration procedure of the instrument to obtain a new calibration constant for the aperture tube. Afterwards, the system suitability check will automatically rerun to check if system suitability is restored. Calibration is triggered only once. If system suitability is not restored after calibration, SystemSuitabilityError is set to True in the protocol and data objects and if AbortOnSystemSuitabilityCheck is set to True, the experiment run will abort without collecting additional data.
    Default Value: Automatic
    Default Calculation: Automatically set to True if any suitability options are specified. Otherwise, set to False.
    Pattern Description: True or False.

    SystemSuitabilityTolerance

    The largest size discrepancy between the measured mean diameter and the manufacture-labelled mean diameter or the measured mean volume and the manufactured-labelled mean volume of SuitabilitySizeStandard sample(s) when sized by electrical resistance measurement to pass the system suitability check. A discrepancy above SystemSuitabilityTolerance triggers the size-calibration procedure of the instrument to obtain a new calibration constant for the aperture tube. Afterwards, the system suitability check will automatically rerun to check if system suitability is restored. Calibration is triggered only once. If system suitability is not restored after calibration, SystemSuitabilityError is set to True in the protocol object and if AbortOnSystemSuitabilityCheck is set to True, the experiment run will abort without collecting additional data.

    SuitabilitySizeStandard

    The particle size standard samples with known mean diameters or volumes used for the checking the system suitability. These standard samples are typically NIST traceable monodisperse polystyrene beads with sizes precharacterized by other standard techniques such as optical microscopy and transmission electron microscopy (TEM). Each of the samples is loaded into the instrument and sized by electrical resistance measurement individually. The system is considered suitable when the measured mean diameter matches the manufacture-labelled mean diameter or the measured mean volume matches the manufacture-labelled mean volume within SystemSuitabilityTolerance.
    Default Value: Automatic
    Default Calculation: Automatically set to a particle size standard sample with a mean diameter closest to 20% of the ApertureDiameter in the catalog if SystemSuitabilityCheck is True. For example, if ApertureDiameter is 100 Micrometer, SuitabilitySizeStandard is automatically set to Model[Sample,"L20 Standard, nominal 20μm, Latex Particle (NIST Traceable), 1 x 15 mL"].

    System Suitability Measurement

    SuitabilityTargetConcentration

    The target particle concentration in the solution from mixing SuitabilitySampleAmount and SuitabilityElectrolyteSampleDilutionVolume in SuitabilityMeasurementContainer. The coincident particle passage effect (multiple particles passing through the aperture of the ApertureTube at approximately the same time to be registered as one larger particle, see ExperimentCoulterCount help file > Possible Issues) becomes severe if SuitabilityTargetConcentration is set too high, leading to a decrease in the accuracy of the measured particle size distribution and measured mean particle diameter or volume.
    Default Value: Automatic
    Default Calculation: Automatically set to the value calculated from the sample concentration, SuitabilitySampleAmount, SuitabilityElectrolytePercentage, and SuitabilityElectrolyteSampleDilutionVolume. If the option information provided is not enough to calculate the target concentration, set to the calculated concentration to dilute the sample concentration by 1000x if SystemSuitabilityCheck is True. If sample concentration is not available, set to Null.

    • Extracting the data for the concetration standard sample, we will use this sample to evaluate the counting capability of the instrument.
    • Extract the DerivedConcentration from the data, which is the calculated concentration of the input sample, from TotalCount, SampleAmount, ElectrolyteSampleDilutionVolumes, and any additional dilutions that may have happened (eg dilution, dynamic dilution options) during the execution of the protocol. Note that the unit is 1/uL.
    • The manufacture concentration is stored in the composition of the model of the sample, in the unit of mol/mL.
    • Convert the manufacture concetration from mol/mL to 1/uL using Avogadros Constant.

    Measure the size distribution and particle count of SamplesIn:

    Sample Dilution

    Sample dilution happens prior to mixing the SamplesIn with ElectrolyteSolution. Specify how the dilution is done for each sample using the "Sample Dilution" options:

    Quantification

    Specify if the concentration of the sample should be updated when the experiment concludes. Only SamplesIn with 1 valid countable analytes (such as cells, polymers etc) in their composition are automatically updated. If SamplesIn contains more than 1 countable analytes, please inspect the data object and manually update SamplesIn using DefineComposition:

    Coincident Particle Passage Effect

    To investigate the best working concentration for SamplesIn, try out different sample loading combinations, and inspect the Oversaturated field of the data and protocol object to determine the best working samples:
    Alternatively, the best working concentration can be investigated using the DilutionStrategy, and serial dilution options:
    Specify if a dynamic dilution process is performed in the sample loading step. In this process, quick preview-type measurement of the SamplesIn-Electrolyte mixture with the initial SampleAmount - ElectrolyteSampleDilutionVolume ratio is performed to check for Oversaturated error. If the error appears, a new diluted sample is prepared by mixing the first mixture with ElectrolyteSolution according to ConstantDynamicDilutionFactor or CumulativeDynamicDilutionFactor and loaded for preview-type measurement again. If the error persists, additional rounds of dilutions are performed until the Oversaturated error disappear, or MaxNumberOfDynamicDilutions of trials is met, whichever one comes earlier:

    System Suitability Check

    Use the SystemSuitabilityCheck option to indicate if you want to check the instrument's sizing capability before measuring the actual input samples, ExperimentCoulterCount will automatically calculate the size standard to use and the corresponding measurement options:

Preferred Input Containers

    Any container that is Aspiratable->True can be used in ExperimentCoulterCount, but the ECL preferred containers (PreferredContainer[All]) are recommended for use in downstream experiments. ExperimentCoulterCount will always pick a new MeasurementContainer, and perform a Transfer from the SamplesIn and ElectrolyteSolution into it for mixing purposes.
    These are the MeasurementContainer option that ExperimentCoulterCount will be set to. Note that if SamplesIn are prepared in one of these containers, ExperimentCoulterCount will still pick another new MeasurementContainer, and perform a Transfer from the SamplesIn and ElectrolyteSolution into the new container for mixing purposes.

Possible Issues

    Coincident Particle Passage Effect

    As the instrument relies on the voltage changes to identify and analyze for one particle passage event, there exists the possibility that two or more particles are passing through the aperture at almost the same time, therefore, giving only one measured signal. This effect is called coincident particle passage effect. The higher the particle concentration, the greater the probability of these multi-passage events will be. This effect can severely downgrade the sizing and counting accuracy if the concentration is too high, and sometimes even block the aperture of the ApertureTube. The concentration of the particles to be measured should be sufficiently low to minimize such effect as much as possible. See "Example Calls/Coincident Particle Passage Effect" section for ways to investigate the best working concentration. If a sample is measured under a concentration where the coincident passage effect is deemed severe by the instrument, the corresponding Field[Oversaturated] will be marked True in the protocol and data object.

    Quantifying Concentration

    The composition of the sample is optionally auto-updated depending on the QuantifyConcentration option value, and number of potential analytes in the input sample after the experiment. If QuantifyConcentration is False, the composition will not be updated. If QuantifyConcentration is True, the composition is updated with the measured value only for the input samples that contain exactly 1 potential analytes (such as cells, polymers etc). Cellular samples will be updated with unit Cell/mL, and other countable analytes will be updated with unit Molar. If input sample contains more than 1 analytes, please inspect the data object stored in the Data field of the protocol, use AnalytePeaks to obtain the count for different analytes/peaks, and update the composition manually using DefineComposition.

    Not Enough MeasurementSample

    Please inspect the calculated SampleAmount, ElectrolyteSampleDilutionVolume, and MeasurementContainers before confirming the protocol to make sure that SampleAmount plus ElectrolyteSampleDilutionVolume will fill up at least 75 percent of max volume of the measurement container model. If the liquid level in the MeasurementContainer is too low, the ApertureTube will not be fully submerged under the liquid, and air bubbles will be introduced into the system when particles are being pumped through the aperture, which will compromise the data quality severely and even prevent the successful completion of the measurement because the air is not electroconductive.

    Volume Mode Recommended

    When calculating particle concentration, the volume difference will measured by either a volume measurement protocol (right before and after the electrical resistance measurement) or the integrated metering pump on the coulter counter instrument. Using the integrated metering pump (i.e. the Volume mode) is always recommended whenever applicable since it provides the best accuracy of determining RunVolume, while a volume measurment protocol may introduce additional error sources including (but not limited to) sample evaporation, balance error (when volume difference is small), and density measurement errors. Volume mode is not available for ApertureDiameter >= 360 Micrometer because there is not enough statistical sampling and Time mode is generally recommended.
