General
MeasurementStrategy
The overall approach used to acquire absorbance readings for SamplesIn. PlateReader holds samples in an open-well container and shines light from above through the liquid column at a pathlength determined by the fill height. Cuvette holds samples in a square container with transparent sides and passes light horizontally at a fixed pathlength. Microfluidic loads samples into narrow channels that gravity-feed measurement chambers read from above at a fixed pathlength. VariablePathlength (Slope Spectroscopy) steps a fibrette optic probe through a range of pathlengths in the same sample and determines concentration from the slope of absorbance versus pathlength, eliminating the need for sample dilution.
Default Calculation: If an Instrument is specified, set to match it (Lunatic to Microfluidic, SoloVPE+ to VariablePathlength, Cary 3500 or another spectrophotometer to Cuvette, any other plate reader to PlateReader). Otherwise set from the specified options: VariablePathlength if any VariablePathlength option is set, Cuvette if SpectralBandwidth or any AcquisitionMix/stirring option is set, PlateReader if any sampling, injection, or plate reader mixing option is set or the samples are prepared robotically, and (when Temperature or EquilibrationTime is set) Cuvette for samples of at least 400 Micro Liter or PlateReader otherwise.
